We’re excited to share insights from our 2022 EU user group meeting on LC-MS analysis of adeno-associated viruses (AAV). This informative session covered an introduction to AAVs, followed by a detailed look at peptide mapping workflows and the identification of post-translational modifications (PTMs) in AAV samples. Understanding PTMs is crucial as they can significantly impact the quality and safety of gene therapy treatments, with two AAV-based therapies already on the market and many more in clinical trials.

The presentation also delved into the intact analysis of AAV samples using advanced techniques. The researchers optimized a workflow employing sodium deoxycholate (STC) and dodecyl maltoside (DDM) for effective peptide digestion and isolation from complex matrices, achieving 100% amino acid sequence coverage in one enzymatic digestion. This comprehensive approach allows for detailed PTM quantification and reveals significant differences between samples. Intact mass analysis confirmed the peptide mapping data, demonstrating that the method does not introduce significant modifications. This webinar provided valuable insights into advanced analytical techniques in gene therapy, making it a must-watch for anyone in the field.
