Easy and fast de novo sequencing of Monoclonal Antibodies

Protein-based therapeutics such as monoclonal antibodies (mAb) are well established in the pipelines of biopharmaceutical organizations and have decades of development ahead. However, not all of them have disclosed or confirmed amino acid sequences.

Liquid chromatography mass spectrometry (LC-MS) provides a powerful tool to obtain the sequence information to the residue level without prior knowledge. Peptide de novo sequencing is the analytical process that determines peptides’ amino acid sequence from their tandem mass spectrum (MS/MS) providing information of the sequence of the whole protein. Data analysis, however, has been challenging until now....

Supernovo™ Workflow

Supernovo is unique in how it determines antibody sequences it mimics the actual biological process of producing antibodies.

The algorithm in silico, converges on a final sequence in an iterative and completely automated process.

The input is raw MS file and the output is the determined sequence with confidence metrics.

Dragonfly Therapeutics

By taking de novo sequencing in house, Dragonfly have improved their turn around time, throughput, their depth of information on their mAbs as well as showing impressive cost savings

Utrecht University

By using a panel of 9 proteases and dual fragmentation Joost Snijder at Utrecht University were able to get the most accurate sequences and the best depth of coverage whilst optimizing protocols for de novo antibody sequencing
