Improving Mass Spectral Quality of Intact Proteins in Complex Mixtures

Presented by Matthew Crawford, Senior Scientist, Rilas Technologies in October 2024

Generating intact protein charge envelopes during mass spectrometry requires careful sample prep and excludes several reagents often used in biological assays. These reagents typically inhibit non-specific interactions between analyte molecules and testing vessels (plates, vials, etc.), while maintaining environments favorable to protein folding. Common examples include salts (NaCl, KCl, MgCl2, phosphate) and detergents (NP-40, SDS, Tween-20), which can suppress signals or complicate spectra.

MOBILion Systems developed a high-resolution ion-mobility system that separates compounds by their collisional cross section (CCS) in the gas phase. This technology separates analytes from contaminants post-ionization, simplifying intact protein analysis. In this seminar, we’ll explore how combining ion-mobility with Protein Metrics HRIM software improves data processing and enhances mass spectral analysis accuracy.
